This is an ELISA (Enzyme Linked ImmunoSorbent Assay) kit for measurement of rat TSH
(thyroid-stimulating hormone) with high sensitivity using Sandwich assay principle.
It is also possible to measure mouse TSH with this kit. The supporting data are shown in the
last part of this instruction.

Thyroid Stimulating Hormone (TSH) is a glycoprotein hormone secreted by
the pituitary gland and regulates the synthesis/ release of T3 and T4 by
thyroid gland. TSH has two subunits, namely alpha and beta. The alpha
subunit is similar to the alpha subunit found in LH, FSH and hCG
glycoprotein hormones. However, the beta subunit is specific and differs
from hormone to hormone.

Approximately 0.2 ml of serum is required per duplicate determination. Collect 4-5 ml of blood into an appropriately labelled tube and allow it to clot. Centrifuge and carefully remove the serum layer. Store at 4oC for up to 24 hours or at - 10oC or lower if the analyses are to be done at a later date. Consider all human specimens as possible biohazardous materials and take appropriate precautions when handling

The determination of serum or plasma levels of thyroid stimulating hormone (TSH or thyrotropin) is recognized as a sensitive method in the diagnosis of primary and secondary hypothyroidism.1 TSH is secreted by the anterior lobe of the pituitary gland and induces the production and release of thyroxine and triiodothyronine from the thyroid gland.2 It is a glycoprotein with a molecular weight of approximately 28,000 daltons, consisting of two chemically different subunits, alpha and beta.3 Although the concentration of TSH in the blood is extremely low, it is essential for the maintenance of nor

The TSH ELISA test is based on the principle of a solid phase
enzyme-linked immunosorbent assay.9,10 The assay system utilizes
a unique monoclonal antibody directed against a distinct antigenic
determinant on the intact TSH molecule. Mouse monoclonal anti-
TSH antibody is used for solid phase immobilization (on the
microtiter wells). A goat anti-TSH antibody is in the antibodyenzyme
(horseradish peroxidase) conjugate solution. The test
sample is allowed to react simultaneously with the two antibodies,
resulting in the TSH molecules being sandwiched between the solid
phase and enzyme-linked ant

The assay is an immunometric ('sandwich') EIA for the quantitative measurement of TSH in human serum or plasma. TSH in the sample is bound by 2 monoclonal anti-TSH antibodies directed at different epitopes. One antibody is attached to magnetic particles and the other is labelled with alkaline phosphatase
