The DNA sample undergoes PCR amplification using primers that flank the specific target region of an HLA locus. This is followed by hybridisation of the resulting amplicon to an array of Sequence Specific Oligonucleotide probes immobilised on a nylon membrane. The SSO probes are designed to hybridise to polymorphic target sequences in the HLA locus. The immobilisation of these probes is referred to as a ‘Reverse Line blot’ and thus gives rise to the acronym ‘RELI™’. Under appropriate wash conditions, the probes remain bound only to their complementary sequence in the amplified DNA. The sensiti

The Biotest ELPHA (Enzyme Linked Probe Hybridization Assay) System is a DNA-hybridization test for the determination of HLA Class I and Class II alleles. Sequence-specific oligonucleotide (SSO-) probes are used as diagnostic tools to identify polymorphic sequence motifs. The hybridization between probe and target DNA is detected by a method adapted from the protein-ELISA technique.

This product is based on the reverse SSO method on a suspension array platform using microspheres as a solid support to immobilize oligonucleotide probes. The target DNA is amplified by polymerase chain reactions (PCR) and then hybridized with the bead probe array. The entire process takes place in a single well of a 96-well PCR plate; thus, 96 samples can be processed at one time.

The LIFECODES HLA typing kits utilize the Luminex xMAP technology and a reverse SSO protocol where the amplified product is hybridized to a mix of beads each coupled with different SSO probes.
Pre-made master mix includes primers, dNTPs and PCR buffer eliminating extra steps and consumables
Homogeneous assay eliminates the need for centrifugation and wash steps saving time and reducing the chances of pipeting errors

LABType® SSO is a reverse SSO (rSSO) DNA typing solution that uses sequence–specific oligonucleotide (SSO) probes and color-coded microspheres to identify HLA alleles. The target DNA is amplified by polymerase chain reactions (PCR) and then hybridized with the bead probe array. The entire process takes place in a single well of a 96-well PCR plate; thus, 96 samples can be processed at one time.
